A meta-analysis of genome-wide association studies of childhood wheezing phenotypes identifies ANXA1 as a susceptibility locus for persistent wheezing

Raquel Granell, John A. Curtin, Sadia Haider, Negusse Tadesse Kitaba et autres

Background: Many genes associated with asthma explain only a fraction of its heritability. Most genome-wide association studies (GWASs) used a broad definition of ‘doctor-diagnosed asthma’, thereby diluting genetic signals by not considering asthma heterogeneity. The objective of our study was to identify …

Author response: A meta-analysis of genome-wide association studies of childhood wheezing phenotypes identifies ANXA1 as a susceptibility locus for persistent wheezing

Raquel Granell, John A. Curtin, Sadia Haider, Negusse Tadesse Kitaba et autres

Using unique data from five longitudinal UK birth cohorts, four distinct subsets of genetic variants were identified as differentially associated across wheezing phenotypes from infancy to adolescence with little evidence of genetic associations spanning across different phenotypes.

Recurrent Severe Preschool Wheeze: From Prespecified Diagnostic Labels to Underlying Endotypes

Polly F. M. Robinson, Sara Fontanella, Sachin Ananth, Aldara Martin Alonso et autres

Abstract Rationale Preschool wheezing is heterogeneous, but the underlying mechanisms are poorly understood. Objectives To investigate lower airway inflammation and infection in preschool children with different clinical diagnoses undergoing elective bronchoscopy and BAL. Methods We recruited 136 children aged 1–5 years (105 …

Circadian asthma airway responses are gated by REV-ERBα

Hannah Jane Durrington, Karolina Krakowiak, P. J. J. De Meijer, Nicola Begley et autres

Background Asthma is an inflammatory disease of the airway showing a strong time of day rhythm. Airway hyperresponsiveness is a dominant feature of asthma, but it is not known if this is under clock control. The circadian clock powerfully regulates inflammation. The …

Pulmonary Group 2 Innate Lymphoid Cell Phenotype Is Context Specific: Determining the Effect of Strain, Location, and Stimuli

Lewis J. Entwistle, Lisa G. Gregory, Robert A. Oliver, William J. Branchett et autres

Group 2 innate lymphoid cells (ILC2s) are enriched at mucosal sites, including the lung, and play a central role in type 2 immunity and maintaining tissue homeostasis. As a result, since their discovery in 2010, research into ILC2s has increased markedly. Numerous …

A T cell–myeloid IL-10 axis regulates pathogenic IFN-γ–dependent immunity in a mouse model of type 2–low asthma

William J. Branchett, Helen Stölting, Robert A. Oliver, Simone A. Walker et autres

Background Although originally defined as a type 2 (T2) immune-mediated condition, non-T2 cytokines, such as IFN-γ and IL-17A, have been implicated in asthma pathogenesis, particularly in patients with severe disease. IL-10 regulates T H cell phenotypes and can dampen T2 immunity to …
