Homeobox transcriptional factor CoHox4 regulates hyphal growth, conidial morphology and appressorial formation in Colletotrichum orbiculare

ホメオボックスは約180 bpから成るホメオドメインと呼ばれるDNA結合モチーフを有する転写制御因子であり,菌類や他の真核生物の形態形成に重要であることが知られている.ホメオボックス転写制御因子をコードする10個のホメオボックス遺伝子がウリ類炭疽病菌のゲノムから同定されている.本研究では,CoHox4遺伝子破壊株を作出し,表現型解析を行うことによって,ウリ類炭疽病菌におけるCoHox4の機能を明らかにした.CoHox4遺伝子破壊株は野生株104-Tと比較して,顕著な菌糸生育能力の低下を示した.培養初期の菌糸生育を観察すると,CoHox4遺伝子破壊株は野生株104-Tと比較して,菌糸の分枝数が多いことが明らかになった.また,CoHox4遺伝破壊株は野生株104-Tと比較して,約1.35倍長い分生胞子を形成した.この細長い分生胞子はガラス上および宿主葉上において付着器形成の異常を示した.さらに,CoHox4遺伝子破壊株は無傷および有傷のキュウリ子葉に対して顕著な病原性の低下を示した.また,CoHox4遺伝子破壊株はカルコフロールホワイトに対する感受性が低かった.以上の結果より,CoHox4が本菌の栄養菌糸生育,分生胞子の形態および付着器形成のような多様な形態分化を制御し,完全な病原性に必要であることが示唆された.

Homeobox transcription factor CoHox2 regulates conidiation and seta formation in Colletotrichum orbiculare

ホメオボックスはホメオドメインと呼ばれる180 bpから成るDNA結合モチーフを有し,転写制御因子をコードする.そして,菌類や他の真核生物では,形態形成に重要な役割を持つことが知られている.本研究では,キュウリ炭疽病の病原体であるウリ類炭疽病菌に保存されている10個のホメオボックスの内の1つであるCoHox2の機能を明らかにした.CoHox2遺伝子を欠損したCoHox2遺伝子破壊株は野生株と比較し,分生胞子形成数が約30%に減少していた.また,CoHox2遺伝子破壊株により形成された分生胞子の形態は野生株104-Tと比較して,大きな差異を示さなかった.さらに,CoHox2遺伝子破壊株はガラス面上で正常に付着器を形成し,宿主植物であるキュウリに病気を引き起こした.また,走査型電子顕微鏡を用いた観察により,剛毛形成数が増加していることが明らかとなった.以上の結果より,CoHox2が分生胞子形成を正に制御し,剛毛形成を負に制御することが示唆された.野生株を用いた顕微鏡観察により,培養7日後に剛毛が形成する分生胞子が細胞核を有することと,培養13日後に剛毛が発芽し,透明な菌糸を伸長していることが明らかとなった.以上の結果より,剛毛が培養初期に細胞核を有する分生胞子を形成する機能と培養後期に菌糸を伸長させる機能を持つことが考えられた.
