MSK1 Downstream Signaling Contributes to Inflammatory Pain in the Superficial Spinal Dorsal Horn

Jahanzaib Irfan, Rizki M. Febrianto, Angelina Mira D’Ercole, N Li et autres

The nuclear kinases mitogen- and stress-activated kinase 1 and 2 (MSK1 and MSK2), through regulating transcriptional processes, are pivotal for various adaptive responses, including inflammation, learning and addiction. Transcriptional alterations in neurons and glia cells within the pain signal-processing (nociceptive) pathway, including …

Mitogen- and stress-activated kinase 1 in primary sensory neurons contributes to formalin-induced tonic pain

Jahanzaib Irfan, Rizki M. Febrianto, Paolo La Montanara, Michelle Meng et autres

Abstract Introduction: The nuclear mitogen- and stress-activated kinases (MSKs) play a critical role in the development and persistence of pain after tissue injury. Objectives: Here, we ascertained the MSK isoform, the cells and mechanisms, which mediate MSKs' pronociceptive function. Methods: Nocifensive behaviour …

Context dependent role of miR-486 promoting neuroregeneration of primary sensory neurons downstream of interleukin-6 signal transducer

Theodora Kalpachidou, Kai K. Kummer, Valentina Handle, David Zimmermann et autres

) show strongly compromised neuro-regeneration, which was significantly rescued by overexpressing miR-486, indicative of a specific role of miR-486 in IL-6/gp130-dependent neuro-regenerative processes. Our findings highlight context-dependent differential expression and roles of miRNAs after nerve injury driving nerve regeneration versus neuropathic pain.

Established and emerging roles of protein kinases in regulating primary sensory neurons in injury-and inflammation-associated pain

David Zimmermann, Michaela Kress, I. Nagy

INTRODUCTION: Recent seminal neuroscience research has significantly increased our knowledge on cellular and molecular responses of various cells in the pain pathway to peripheral nerve injuries and inflammatory processes. Transcriptomic and epigenetic analysis of primary sensory neurons (PSNs) in animal models of …

Biophysical essentials – A full stack open-source software framework for conserved and advanced analysis of patch-clamp recordings

David Zimmermann, Michaela Kress, Maximilian Zeidler

BACKGROUND AND OBJECTIVES: Patch-Clamp recordings allow for in depth electrophysiological characterization of single cells, their general biophysical properties as well as characteristics of voltage- and ligand-gated ionic currents. Different acquisition modes, such as whole-cell patch-clamp recordings in the current or voltage clamp …

Novel Metric for Non-Invasive Beat-to-Beat Blood Pressure Measurements Demonstrates Physiological Blood Pressure Fluctuations during Pregnancy

David Zimmermann, Hagen Malberg, Martin Schmidt

Beat-to-beat (B2B) variability in biomedical signals has been shown to have high diagnostic power in the treatment of various cardiovascular and autonomic disorders. In recent years, new techniques and devices have been developed to enable non-invasive blood pressure (BP) measurements. In this …

IL-4 shaping glutamatergic synapse like structures for more mature iPSC derived neuron phenotypes

David Zimmermann, Clemens L. Schöpf, Georg Kern, Theodora Kalpachidou et autres

Abstract The prototypical pleiotropic anti-inflammatory cytokine IL-4 not only acts on immune cells but also has important roles in the nervous system mediating antinociception and neuroregeneration. Therefore, we explored the expression of IL-4, its receptors IL4RA and IL13RA1 and downstream signaling components …
