This archive includes: Data: raw data, processing code, and processed data Hardware: parts list, design files, and assembly instructions Software: compiled JAR file and source code Article Abstract: Polarization differential phase contrast microscopy (pDPC) is a robust, low-cost single-shot implementation of quantitative phase imaging based on differential phase contrast (DPC) microscopy utilising a polarization-sensitive camera to simultaneously acquire four obliquely illuminated images. We recently reported a cost-effective open-source implementation of pDPC on a modular “openFrame-based” microscope, where the spatial resolution was limited by the numerical aperture (NA) of the condenser lens, as is generally the case for condenser-based DPC. Here we present even lower-cost open-source implementations that overcome this limitation by using domestic lighting illumination sources to realise quantitative phase imaging (QPI) at up to ~0.95 NA. We have applied these pDPC implementations to image mammalian cells and mycobacteria.
