MaaT013 fecal microbiotherapy modulates key gastro-intestinal and immunological processes involved in GvHD pathogenesis in vitro and in vivo

The purpose of the study was to evaluate the mechanism of action of MaaT013 fecal microbiotherapy in GI-aGvHD. Here, we used multi-omics analyses in complementary in vitro and in vivo models.In an in vitro, inflammation-induced leaky- model, MaaT013-derived metabolites prevented barrier disruption.In vitro, MaaT013 promoted dendritic cell-mediated expansion of Tregs and stimulated IL-10 and TGF-b secrection. In germ-free mice, MaaT013-derived beneficial bacteria engrafted, produced anti-inflammatory metabolites, and restored physiology through immune rebalancing.In a humanized mouse model, MaaT013 delayed onset of GvHD symptoms.All data were generated using standardized experimental procedures between 2020 and 2025.Overall, these findings show that MaaT013 can counteract key pathways of GI-aGvHD pathogenesis by restoring microbial diversity, reinforcing intergrity, and re-establishing immune homeostasis.
