Progress in cancer immunotherapy and immunogenicity optimization strategies based on tumor-specific antigens

Tumor-specific antigen including tumor neoantigens, which are antigens expressed only on the surface of tumor cells and not on normal cells, can stimulate the body's anti-tumor immune response and have great potential for personalized precision treatment. This article reviews the types, screening and prediction methods of tumor neoantigens, and their application effects in non-cellular therapy, cellular immunotherapy and combination therapy, and summarizes the current optimization strategies for neoantigens to enhance immunogenicity and achieve efficient delivery. Immunotherapy based on neoantigens is expected to become a key force in overcoming tumor problems and provide new hope for personalized cancer treatment.
