Comparative effectiveness of antidepressants and antipsychotics for management of opioid use disorder: a systematic review and network meta-analysis of randomized trials

Opioid use disorder (OUD) is a chronic, relapsing condition characterized by compulsive substance use despite negative consequences. OUD is associated with significant psychiatric comorbidities, including depression, anxiety, and psychotic symptoms, which are often managed with antidepressant and antipsychotic medications. However, the effectiveness and safety of these medications in OUD populations remain uncertain, contributing to inconsistent clinical practices and prescribing patterns that may not be based on strong evidence. This systematic review and network meta-analysis will assess the comparative effectiveness and safety of antidepressants and antipsychotics in management of OUD. By synthesizing the available evidence, this review aims to clarify treatment outcomes, identify research gaps, and support evidence-based clinical guidelines for management of OUD.
