Real-time optogenetic control of excitation wavefronts in human cardiomyocytes using holographic one- and two-photon stimulation

An all-optical method to monitor, stimulate and feedback-control excitation wavefronts in human cardiomyocyte networks in real-time is presented. We employ ferro-electric spatial light modulators for fast (~kHz) display of excitation patterns and sparsely sampled optical signals to characterize the prevalent state of the excitation wavefront. Experiments were conducted on human-induced pluripotent stem cell-derived cardiomyocytes expressing the opsin Chrimson. We demonstrate that in the case of wavefront distortions the closed-loop control can restore the prevalent wavefront form successfully automatically and in real-time. The presented all-optical approach paves the way for label-free investigation of optogenetic real-time closed-loop cardioversion strategies.
