Essential oil analysis and isolation of bioactive compounds from the leaf of Myrica salicifolia: Computational pharmacokinetics and toxicity assessment

This study investigates the chemical composition of essential oils and isolates bioactive compounds from the leaves of Myrica salicifolia, a plant used in traditional Ethiopian medicine. Gas Chromatography-Mass Spectrometry (GC-MS) analysis of the essential oils of the leaves of the plant resulted in the identification of 21 major volatile components, with trans-3-hexen-1-ol (52.62%), terpineol (7.93%), and benzaldehyde (6.28%) as predominant compounds. Phytochemical analysis of a methanol-dichloromethane (1:1) extract of the leaf of M. salicifolia led to the isolation of four compounds (1 – 4). All of these compounds are reported for the first time from the M. salicifolia. Structural elucidation was performed using their NMR, FT-IR, and HR-MS analysis. Computational analyses were conducted using Swiss ADME for pharmacokinetic predictions and ProTox-II for toxicity assessment. Lipinski's rule of five and other ADME parameters were applied to evaluate oral bioavailability and drug-likeness. Results showed that compounds 1 and 4 complied with Lipinski's rules, exhibited good oral bioavailability, and showed low toxicity, making them promising candidates for drug development.
