Rentosertib, a generative AI-discovered TNIK inhibitor improves lung function in IPF patients

Rentosertib, a novel AI-generated TNIK inhibitor, was tested in a 12-week phase 2a randomized, double-blind, placebo-controlled trial of 71 idiopathic pulmonary fibrosis (IPF) patients. Participants received placebo, 30mg once daily (QD), 30mg twice daily (BID), or 60mg QD. Baseline demographics and lung function were comparable across groups and the rate of TEAES were similar across all treatment groups. At 12 weeks, placebo-treated patients showed a mean forced vital capacity (FVC) decline of -62.3mL (95% CI: -115.3 to -9.33). The 30mg QD group had reduced decline (-37.5mL; -100.8 to 25.7), while 30mg BID and 60mg QD groups showed FVC improvements: +19.7mL (-60.5 to 99.9) and +98.4mL (10.9 to 185.9), respectively. Rentosertib-treated patients maintained FVC benefits regardless of antifibrotic use: +25.6mL (-58.9 to 110.1) without antifibrotics vs. placebo’s -54.5mL (-119.3 to 10.3), and +20.3mL (-32.5 to 73.1) with antifibrotics vs. placebo’s -79.9mL (-183.3 to 23.5). This study highlights Rentosertib’s potential to stabilize lung function in IPF via anti-fibrotic/anti-inflammatory effects and demonstrates a significant advance in AI-driven drug discovery, accelerating therapeutic development.
