Caproic acid-producing bacteria (CPBs) are considered of great importance to the aging of pitmud within the Chinese strong-flavor Baijiu ecosystem. However, the role of CPBs during pitmud aging remains unclear. To address this question, autochthonous CPBs were subjected to successive liquid-state enrichment and solid-state pre-cultivation with fresh bottom pitmud, followed by five consecutive batches of fermentation to artificially accelerate pitmud aging. This study revealed how CPBs and related prokaryotic communities evolved during these stages. The enriched CPBs did not grow as rapidly in the solid-state new pitmud as in the liquid state. The multi-dimensional succession pattern of bottom pitmud prokaryotic communities, α-diversities, β-diversities, physicochemical properties, and the predicted caproic acid synthesis functions co-verified the "Lag-aging dual-phase" hypothesis proposed in this study. This hypothesis divides pitmud aging into two distinct phases: a "Lag phase" (adaptive phase) and an "aging phase". After five fermentation batches, the prokaryotic community structure of the new bottom pitmud closely resembled that of aged old bottom pitmud, and the CPBs gradually concentrated in the surface layer (0-1 cm). This study therefore confirmed that the inoculation with autochthonous CPBs-rich consortia facilitated pitmud aging, paving the way for artificial pitmud aging and targeted bioremediation.
