Ketoconazole-loaded nanoemulsion in situ gel system for ocular delivery: development and evaluation

This study deals with the development of nanoemulsion containing ketoconazole loaded in situ gel system for ocular delivery, followed by in vitro and ex vivo, evaluations. Nanoemulsion formulation was characterized by globular size, polydispersity index, pH, zeta potential, and drug content. In situ, gel systems based on Pluronic and xyloglucan exhibited temperature-responsive gelation. The nanoemulsions exhibited a droplet size are 192.8 nm with a PDI value of 0.362. The in situ gel formulation was found to be isotonic and exhibits sustained release for 6 h. There is better antifungal activity of the in situ gel formulation (32.58 ± 2.13 nm) than the marketed eye drop (30.23 ± 1.74 nm). The observations of the HET-CAM test showed that the developed in situ gel formulation is essentially non-irritating and possesses good ocular tolerability. Conclusively, a nanoemulsion-based in situ gel system is a promising approach to delivering ketoconazole for enhanced permeation and therapeutic efficacy after ocular administration.
