Study on the Detection Method of Rheumatoid Factor Based on Nano-Microspheres

Rheumatoid factor (RF) plays an important role in clinical practice as an important indicator for detecting immunologic diseases. The aim of this study was to develop a system for the detection of rheumatoid factor (RF) using nanomicrosphere immunoturbidimetric assay (PETIA). This study will optimize the traditional method of reagent preparation by coupling small-sized microspheres to antigen to extend the linear range of the assay and adding large-sized microspheres to increase the sensitivity of the assay and make the kit more accurate. Methods: Screening (RF antigen, particle size of nano-microspheres) and determining appropriate reaction conditions, combining RF antigen with nanomicrospheres to establish PETIA detection of RF, and finally performance evaluation and analysis. RESULTS: The reagents were prepared by using RF antigen, small-size nanomicrospheres JSR-P0115 and large-size nanomicrospheres JSR-P0323 from Aoyan Bio, and the ratio of the small spheres to the large spheres was 10:1. The relative deviation of the accuracy of the method was not more than 2%; the CV value of the inter- and intra-batch precision was not more than 3%; and the linear range was from 0 to 390 mg/L, with the linear equations of y= 0.0017x + 0.0023, R2=0.9979 > 0.99, which is better than the commercially available kits. The reagent is easy to use, with accurate results, wide linear range, good stability, low price, and can be completely used for the detection of clinical specimens.
