Multiple sclerosis genetics in an ancestrally-diverse UK population

Background Genome-wide association studies (GWAS) of Multiple Sclerosis (MS) susceptibility and severity have primarily focussed on populations of European ancestry. Studying MS genetics in other ancestral groups is necessary to ensure the generalisability of these findings. Methods ADAMS is a nationwide multi-centre genetic study of MS susceptibility and severity focussing on individuals with MS from non-European ancestral backgrounds. Participants are recruited via participa- ting clinical sites, the website (https://app.mantal.co.uk/adams), or the UK MS Register. We are collecting phenotypic data and genotyping participants using saliva. Results We have recruited 682 participants within the first year of the study (71.2% female, median age 44.9). Over 60% of the cohort are non-White, with 23.5% identifying as Asian, 16.2% as Black, and 20.9% identifying as having mixed or other backgrounds. Preliminary genetic analyses confirm the ancestral diversity of the cohort and recapitulate the role of Major Histocompatibility Complex (MHC) alleles in determining MS risk. Conclusions ADAMS is an ongoing genetic study of MS susceptibility and severity in an ancestrally-diverse cohort. In the long-term, genetic data will be combined with other datasets to further cross-ancestry genetic discoveries.
