Funding Acknowledgements: Supported by grants from the British Heart Foundation (to JK and MA) and NIH (to BJB and YG) Introduction: The positive inotropic and lusitropic effects of β-adrenoceptor stimulation are mediated in part by protein kinase A (PKA)-mediated phosphorylation of cardiac troponin-I (cTnI), which decreases myofibrillar Ca2+ sensitivity, and of cardiac myosin-binding protein C (cMyBP-C), which accelerates myosin crossbridge kinetics. Previous evidence suggests that phosphorylation of both Ser23 and Ser24 in cTnI is required for the Ca2+ desensitization by PKA. Phosphorylation by PKA also protects cTnI from proteolysis by calpain. Here we report that, unlike PKA, protein kinase D (PKD) phosphorylates cTnI at only one site in Ser23/24. We examined the functional consequences of this monophosphorylation in mouse cardiac myofibrils. Purpose: To compare myofibrillar Ca2+ sensitivity and susceptibility of cTnI to calpain-mediated proteolysis when Ser23/24 sites in sarcomeric cTnI are unphosphorylated, monophosphorylated or bisphosphorylated. Methods: Detergent-skinned ventricular trabeculae from mouse were dephosphorylated using λ–phosphatase. Ca2+-activated force and crossbridge kinetics (ktr) were measured before and after muscle incubation with PKD, then again after PKA. Isolated myofibrils were treated similarly for measurement of cTnI and cMyBP-C phosphorylation. Myofibrils were also treated with calpain and the degradation of cTnI and cMyBP-C was measured.
