Characterization of prostaglandins by LC/APCI-MS.

LC/APCI-MS(liquid chromatography/atmospheric pressure chemical ionization-mass spectrometry)を用いて,プロスタグランジン類(PGs;PGA1,PGB1,PGD2,PGE1,PGE2,6-keto-PGF1α)の正イオンモードでの同時測定を行った.マススペクトルは主にPGsから水が脱離した脱水体イオンが基準イオンピークとして検出され,移動相中にAcONH4を添加することによりPGB1以外のPGsはNH4付加イオンが基準イオンピークとして検出された.NH4付加体生成の可否はPGsの立体分子構造及び表面の荷電状態に関連しているものと考えられた.カラム分離したときのTIC(total ion current)モードでの検出限界は約100ngであった.又,SIM(selected ion monitoring)モードでの検量線の相関係数はいずれも0.998以上と良好であり,検出限界は2～6ng,同一試料溶液を5回注入したときの再現性は,相対標準偏差で約5%であった.この結果LC/APCI-MSにより,PGsの同時定量が可能であることが確認された.
