肥満を伴った認知症発症に対するコーヒーの予防効果の可能性

近年,肥満がアルツハイマー型認知症(AD)の危険因子であるというエビデンスが集積されてきた。一方,脳由来神経栄養因子(BDNF)は神経細胞の維持,修復など重要な働きをするタンパク質であるが,脳内でのBDNF発現量がAD患者で減少していることが確認されている。これまで我々は,小腸から分泌されるGLP-1が門脈を経由して肝臓内のGLP-1受容体に作用し,さらに肝臓からの求心性神経を介して脳内BDNF発現を高めるということを明らかにしている。さらに肥満モデル動物において,GLP-1の発現が低下することも認めている。今回肥満モデル動物を用い,コーヒー摂取が小腸内GLP-1の発現を増加させ,それに伴い脳内BDNF発現も増加させるかを検討した。コーヒーを摂取すると,肥満による小腸内の炎症性変化が抑制され,小腸内GLP-1発現の低下も改善した。さらにコーヒー摂取は,肥満による脳内BDNF発現の低下も抑制させた。コーヒー摂取は認知症の発症予防に有効である可能性が示唆された。
