tinydenseR: Linking Cell-To-Cell Variation to Sample-to-Sample Variation

Pedro Milanez‐Almeida, Daniela Schildknecht, Markus Linder, Saskia M. Brachmann et autres

tinydenseR is a landmark-based platform for single cell data analysis that identifies differential cell states and features, including subtle within-cluster state changes. Modeling samples as replicates, tinydenseR enhances analytic efficiency and reproducibility while preserving the richness of single cell data.

Sample-level modeling of single-cell data at scale with tinydenseR

Pedro Milanez‐Almeida, Daniela Schildknecht, Markus Linder, Saskia M. Brachmann et autres

Abstract Single-cell studies now routinely encompass hundreds of samples and millions of cells, offering unprecedented opportunities to link sample-level phenotypes with cellular and molecular states. However, current workflows often depend on cell-level inference and rigid clustering, which can distort significance and obscure …

Data from CRISPR Screening Identifies Mechanisms of Resistance to KRASG12C and SHP2 Inhibitor Combinations in Non–Small Cell Lung Cancer

Anirudh Prahallad, Andreas Weiss, Hans Voshol, Gráinne Kerr et autres

Abstract Although KRASG12C inhibitors show clinical activity in patients with KRAS G12C mutated non–small cell lung cancer (NSCLC) and other solid tumor malignancies, response is limited by multiple mechanisms of resistance. The KRASG12C inhibitor JDQ443 shows enhanced preclinical antitumor activity combined with …

Data from FGF401, A First-In-Class Highly Selective and Potent FGFR4 Inhibitor for the Treatment of FGF19-Driven Hepatocellular Cancer

Andreas Jeffrey Weiss, Flavia Adler, Alexandra Buhles, Christelle Stamm et autres

Abstract Hepatocellular carcinoma (HCC) is the most common primary malignancy of the liver and it is the third leading cause of cancer-related deaths worldwide. Recently, aberrant signaling through the FGF19/FGFR4 axis has been implicated in HCC. Here, we describe the development of …
