Evolution mechanism of bacterial drug resistance in aquatic environments driven by typical epidemic control drugs

抗生素耐药性已成为全球重大健康威胁,而抗生素的过度使用是驱动细菌耐药性发展的关键因素。新冠疫情期间,大量控疫药物进入水环境并被频繁检出,然而其对环境水体中微生物抗生素耐药性的影响尚不明确。为此,本研究选取典型控疫药物磷酸氯喹和磷酸奥司他韦,对耐多粘菌素大肠杆菌E. coli DH5α (MCR)进行迭代暴露实验,以明晰其对细菌耐药性演化的作用机制。结果发现:随着2种控疫药物暴露浓度的增加,细菌胞内活性氧物种(ROSs)水平呈现先升后降的趋势,同时超氧化歧化酶活性(SOD)与过氧化氢酶(CAT)活性变化趋势与ROSs基本一致,表明氧化应激响应被激活。此外乳酸脱氢酶(LDH)释放量持续上升,细胞内外膜通透性先增强后趋于减缓。菌体形态出现明显变化,其中磷酸氯喹暴露组细菌个体尺寸显著缩短。在持续药物诱导下,细菌生长活性受到持续抑制,发育过程受阻,代谢活性逐步下降。10、100、1000 mg/L浓度暴露均促使耐多粘菌素菌株不仅对多粘菌素耐药性进一步增强,还广泛产生对其他抗生素的交叉耐药。经100 mg/L磷酸氯喹传代80代后,菌株对多粘菌素、头孢噻肟、美罗培南、环丙沙星、红霉素、四环素的耐药性分别提高至16、10、5、10、3和15倍;而相同浓度磷酸奥司他韦暴露亦导致相应耐药水平提升16、15、5、8、3和15倍。由此可见,水体细菌在典型控疫药物暴露下可显著增强其适应性与耐药演化潜力,并可发展为多重耐药表型。本研究从微生物传代的角度揭示了控疫药物促进耐药性形成的生理与氧化应激机制,可为未来控疫药物管控与耐药性防控提供了新的视角。
