Regulation of TRAIL-induced Cancer Cell Death by Fucosylated Glycans

フコシル化糖鎖は、一般的にがん化に伴い増加する糖鎖構造として知られ、フコシル化分子は腫瘍マーカーとして利用されてきた。フコシル化糖鎖の機能は多岐に渡っているが、がんの増殖や転移に関わることが知られている。フコシル化糖鎖を合成するにはフコース転移酵素のドナー基質となるGDP-フコースが必須であるが、我々はGDP-フコース合成に必須の酵素であるGDP-mannose 4,6-dehydratase(GMDS)遺伝子のがんにおける異常とそれによるGMDSタンパク質の欠損を世界に先駆けて報告してきた。また、このGMDS遺伝子異常の発見を契機に、フコシル化糖鎖が細胞死誘導分子Tumor necrosis factor-related apoptosis-inducing ligand(TRAIL)および腫瘍免疫回避機構への感受性を制御していることを明らかにしてきた。本総説では、我々のこれまでの知見を中心に、フコシル化糖鎖によるがん細胞死の制御について概説したい。
