Effects of Glove Materials on Porcine Sperm Motility and Acrosome Morphology

ゴム製手袋はブタ精子の性状に悪影響を及ぼすことが指摘されているが,現在もなおゴム製手袋がブタの精液採取時に使用されている。ゴム製品は,多くの成分から形成されており,製品によって精子に与える影響が異なる可能性がある。本研究では,現在流通しているゴム製品を含めた使い捨て手袋の材質が精子性状に与える影響について,わが国で養豚用に飼育されている一般品種を用いて検証した。実験1では,生ゴム,ニトリルゴム,加硫促進剤不使用のニトリルゴムおよびポリエチレン製手袋の断片と精液を共培養した。精子の運動性は,生ゴム,ニトリルゴムおよび加硫促進剤不使用のニトリルゴムでは著しく低下した。一方,ポリエチレンは共培養しないコントロールと差がなかった。先体の形態に与える害作用は運動性に比較すると軽度ではあったが,ニトリルゴムおよび加硫促進剤不使用のニトリルゴムでは,培養時間が長くなるにつれて増加した。実験2では,ニトリルゴムを水あるいは石鹸で十分に洗浄し,精液と共培養した。洗浄処理は精子の運動性や先体正常性に対する害作用を一定程度減少させたが,その効果は不十分なものであり,洗浄後も共培養した精子の運動性および先体正常性は共培養しないコントロールに比べて低い値を示した。実験3では,ポリエチレン製手袋4種類と精液を共培養した。1種類のポリエチレン製手袋において,運動性および先体正常性が低下したが,残り3種類はコントロールと差がなかった。これらの結果から,生ゴムおよび加硫促進剤不使用を含めたニトリル製ゴム製手袋は,ブタ精子の性状,特に運動性に対して著しい害作用を示すこと,この害作用は洗浄処理では十分に除去できないこと,ポリエチレン製手袋の多くは安全性が非常に高いことが明らかとなった。したがって,精液の採取や処理を行うときにはゴム製品の使用をやめ,ポリエチレン製品など安全性が高いものを使用することが推奨される。
