AI-2 production in Fusobacterium nucleatum is subspecies-specific and uncoupled from quorum sensing

Bibek G C, Shiqi Xu, Truman Tran, Chenggang Wu

ABSTRACT Autoinducer-2 (AI-2) is a LuxS-dependent product of the activated methyl cycle (AMC) that functions as a quorum-sensing signal in diverse bacteria. Fusobacterium nucleatum is a genetically heterogeneous oral anaerobe comprising four subspecies: nucleatum (FNN), vincentii (FNV), polymorphum (FNP), and animalis (FNA). …

Complete genome sequence of the widely used coaggregation fusobacterial strain PK1594

Bibek G C, Shiqi Xu, Chenggang Wu

ABSTRACT We report the complete genome sequence of the fusobacterial strain PK1594, widely used to study coaggregation between Fusobacterium nucleatum and oral bacteria. The assembly comprises a 2,357,192-bp chromosome and a 20,441-bp contig representing a duplicated 10,221-bp plasmid. znpA analysis indicates that …

The gene encoding ornithine decarboxylase for putrescine biosynthesis is essential for the viability of Fusobacterium nucleatum

Shiqi Xu, Bibek G C, Alex Phan, Chenggang Wu

ABSTRACT Fusobacterium nucleatum is a Gram-negative anaerobe associated with periodontitis and colorectal cancer. It secretes putrescine, a polyamine that promotes biofilm formation by oral co-colonizers and enhances the proliferation of cancer cells. However, the physiological importance of putrescine for F. nucleatum itself …

Fusobacterium nucleatum produces previously unappreciated AHR-activating metabolites and promotes CRC cell proliferation via the AHR-TERT axis

Kyoung‐Jae Won, Pei-Ru Jin, Lydia H. Davis, Jimmy Orjala et autres

SUMMARY Fusobacterium nucleatum ( Fn ) is prevalently enriched in colorectal cancer (CRC), promoting CRC progression. However, Fn -derived small molecules and their host target pathways in CRC remain largely underexplored. Here, we identify that Fn produces previously unrecognized three indole-containing metabolites, …

Complete genome sequence of Actinomyces oris strain MG-1: an oral commensal critical for dental plaque formation

Bibek G C, Kexin Tan, Chenggang Wu

MG-1 is a key oral commensal involved in dental plaque formation. We report its complete genome sequence, generated using Illumina and Oxford Nanopore sequencing. The genome consists of a single circular chromosome of 3,042,829 bp, providing a resource for future functional studies.

Inactivation of the Fusobacterium nucleatum Rnf complex reduces FadA-mediated amyloid formation and tumor development

Timmie A. Britton, Ju Huck Lee, Chungyu Chang, Aadil Hussain Bhat et autres

ABSTRACT The Gram-negative anaerobe Fusobacterium nucleatum is an oral oncobacterium that promotes colorectal cancer (CRC) development with the amyloid-forming cell surface adhesin FadA integral to CRC tumorigenesis. We describe here molecular genetic studies uncovering a novel mode of metabolic regulation of FadA-mediated …

The CarSR two-component system directly controls radD expression as a global regulator that senses bacterial coaggregation in Fusobacterium nucleatum

Bibek G C, Chenggang Wu

ABSTRACT Two-component systems (TCS) enable bacteria to sense and respond to environmental signals, facilitating rapid adaptation. Fusobacterium nucleatum , a key oral pathobiont, employs the CarSR TCS to modulate coaggregation with various gram-positive partners by regulating the expression of radD , encoding …
