Lettuce ( Lactuca sativa ) laticifers are specialized cells that synthesize high-quality natural rubber, serving as an accessible model for latex metabolism. However, the global metabolic landscape and regulatory circuitry within laticifers remain largely unexplored. Here, we performed multi-tissue transcriptomics, enzyme activity profiling, and targeted metabolite quantification to systematically decipher the functional architecture of lettuce laticifers. Our integrative analysis suggests that laticifers operate as a multifunctional metabolic hub, where mevalonate (MVA)-driven rubber biosynthesis co-occurs with highly active ascorbate (AsA) synthesis and regeneration pathways. This spatial convergence not only supports rubber production but also may establish a redox buffering system that could confer protection against oxidative stress, linking specialized metabolism to stress defense. Furthermore, we find that the ubiquitin-proteasome system (UPS) and a MYC2-centered jasmonate (JA) / abscisic acid (ABA) signaling module are associated with this hub. Rather than presenting a mere descriptive catalog of differentially expressed genes, our work points to a coherent regulatory and metabolic network with specific nodes (e.g., HMGCR , GGP , DHAR , MYC2 , UBE2D ) that may serve as candidates for metabolic engineering. These findings transform lettuce laticifers from an underexplored system into a tractable platform for improving rubber production, stress resilience, and nutritional value in horticultural crops.
