The Expression of Long Noncoding RNA FOXD2-AS1 and OIP5-AS1 in Breast Cancer and Its Relationship with Clinicopathological Characteristics

Background: Breast cancer is one of the most common malignancies in women. A major challenge in its diagnosis and treatment is the lack of adequate molecular targets. Various risk factors can influence the likelihood of developing breast cancer. Reduced expression of long noncoding RNAs (lncRNAs), such as OIP5-AS1 and FOXD2-AS1, has been associated with carcinogenicity, patient survival, prognosis, and the progression of several types of cancer. Objectives: Given the current lack of molecular tests for accurate breast cancer diagnosis, this study aims to investigate whether OIP5-AS1 and FOXD2-AS1 can serve as diagnostic biomarkers for breast cancer. Methods: This study included 50 women diagnosed with breast cancer who had not yet received any treatment. Paired tumor and adjacent normal tissue samples were collected from each patient. Total RNA was extracted using the RiboEx Total RNA Solution. Complementary DNA (cDNA) was synthesized using the ExcelRT Reverse Transcription Kit. The expression levels of FOXD2-AS1 and OIP5-AS1 were measured by quantitative real-time PCR (qRT-PCR), and gene expression data were normalized using the 2−ΔΔCt method. Appropriate statistical tests were applied to compare gene expression levels between tumor and normal tissue samples. Results: This study identified a significant association between lncRNA FOXD2-AS1 expression levels and lymph node metastasis in patients with breast cancer. In addition, increased expression levels of both lncRNAs FOXD2-AS1 and OIP5-AS1 were significantly correlated with several clinical and demographic factors, including age, menopausal status, the presence of underlying conditions, history of chest radiography, and stress and sleep patterns. Conclusions: The elevated expression of lncRNAs FOXD2-AS1 and OIP5-AS1 in breast cancer tissues underscores their potential as biomarkers for disease progression.
