Responses of the pine wood nematode (Bursaphelenchus xylophilus) and host pine trees (Pinus massoniana) to exogenous calcium application

The pine wood nematode (PWN), Bursaphelenchus xylophilus , is one of the most serious pest and disease threats facing Pinus massoniana , severely limiting its current economic value; therefore, there is an urgent need to find new approaches to address this situation. The application of exogenous calcium can enhance the plant’s stress tolerance and influence various physiological processes in P . massoniana through a series of biotic and abiotic interactions. In this study, two application methods were used to investigate the resistance of P . massoniana to B . xylophilus under exogenous calcium application, as well as the physiological and biochemical responses of the trees. The results showed that under exogenous calcium application (10 mmol/L and 50 mmol/L), P . massoniana effectively reduced the population growth and migration rate of B . xylophilus within the tree when responding to nematode invasion, preliminarily confirming that exogenous calcium application can enhance the resistance of P . massoniana to B . xylophilus . Calcium ion absorption by different parts of P . massoniana varied with the application method. Under calcium application, the activities of antioxidant enzymes (PPO, POD, PAL, and CAT) in P . massoniana increased; however, the extent of the increase in each enzyme activity differed among the application methods, indicating differences in the defense responses of P . massoniana under different calcium application methods. These results suggest that exogenous calcium activates the defense enzyme system, thereby inhibiting nematode proliferation and migration within the tree and enhancing the disease resistance of P . massoniana .
