マウスES細胞を用いた全能性細胞誘導法及びブラストイド生成法の開発

本研究の目的は、マウスES細胞から人工的に全能性様状態を誘導し、胚体外系列を含むすべての胚構成細胞系列を試験管内で再構成する技術を確立することである。近年、ES細胞からごく低頻度で自発的に出現する2細胞期様細胞(2CLC)が全能性様の性質を示すことが報告されており、その誘導メカニズムの解明は幹細胞生物学における重要な課題である。申請者は、先行研究においてscRNA-seq解析を用いて2CLCに特異的な転写因子を探索し、レチノイン酸シグナルに関与するRXRαおよびHmgn3を同定した。これを受けて、Hmgn3の発現誘導によって全能性様状態が再現できるのではないかとの仮説を立てた。 本年度はまず、ドキシサイクリン依存的にHmgn3を発現誘導できるES細胞株を樹立した。この細胞株を用いて、3D培養環境下でブラストイドと呼ばれる人工胚盤胞モデルの形成条件を系統的に検討し、高効率でブラストイドが形成される培養条件を確立した。得られたブラストイドは、免疫染色解析により、Cdx2陽性のTE(栄養外胚葉)系統およびOct4陽性のICM(内部細胞塊)系統への明確な分化を示し、自然な胚盤胞に類似した細胞系譜構成を有することが確認された。さらに、これらのブラストイドからトロフォブラスト幹細胞(TSC)の樹立にも成功し、Hmgn3の発現誘導によってES細胞が全能性様状態を経て胎盤系列幹細胞へと分化可能であることが示唆された。ES細胞から胎盤系列細胞を誘導する技術は、再生医療や不妊症研究への応用につながると期待される。以上の成果は、申請時に掲げた「全能性細胞の誘導法およびブラストイド生成法の開発」という目標を達成するものであり、ES細胞の可塑性や全能性獲得メカニズムの理解に貢献することが期待される。 The objective of this study was to establish a technique for artificially inducing a totipotent-like state from mouse embryonic stem (ES) cells and reconstructing all embryonic cell lineages, including extraembryonic tissues, in vitro. In recent years, it has been reported that a very small fraction of ES cells spontaneously convert into two-cell-like cells (2CLCs), which exhibit totipotent-like properties. Elucidating the mechanisms underlying this conversion remains a key issue in stem cell biology. In our previous work, we performed single-cell RNA sequencing (scRNA-seq) to identify transcription factors specifically expressed in 2CLCs, and identified RXRα and Hmgn3, both of which are involved in retinoic acid signaling. Based on these findings, we hypothesized that induction of Hmgn3 expression could recapitulate the totipotent-like state. In the current year, we established a doxycycline-inducible ES cell line capable of Hmgn3 overexpression. Using this cell line, we systematically screened culture conditions for blastoid formation under three-dimensional (3D) culture environments and successfully identified conditions that enabled efficient generation of blastoids. Immunofluorescence analysis revealed that the resulting blastoids displayed distinct lineage specification: Cdx2-positive trophectoderm (TE) and Oct4-positive inner cell mass (ICM), recapitulating the lineage composition observed in natural blastocysts. Furthermore, we succeeded in deriving trophoblast stem cells (TSCs) from these blastoids, indicating that ES cells can differentiate into extraembryonic lineages via a totipotent-like intermediate state upon Hmgn3 induction. The ability to generate trophoblast lineage cells from ES cells without genetic modification holds promise for applications in regenerative medicine and reproductive biology. These findings fulfill the research goal of developing a method to induce totipotent-like cells and generate blastoids, and represent a significant advancement in our understanding of ES cell plasticity and the mechanisms underlying totipotency acquisition.
