Chemically induced mutagenesis of a new pea variety Pamyati Popova using colchicine

The article looks at the effect of treating seeds of the pea variety Pamyati Popova with colchicine on their morphobiological traits. The purpose of the study is to determine the optimal conditions for using colchicine to induce polyploidy in field peas and to assess the morphobiological and economic-value traits of the resulting lines. The study's novelty lies in its determination, for the first time, of the appropriate colchicine concentration and the duration of seed treatment for a new pea variety under the conditions of the Republic of Bashkortostan, which are crucial for the successful induction of polyploidy. The highest values for the number of seeds per plant (17.8 ± 0.2 psc.) and seed weight (45.3 ± 0.4 psc.) were observed in mutant plants treated with a 5 mg/l colchicine solution for 24 hours, which was statistically significantly higher than the control. Histological studies of the mutants showed that the number of chloroplasts in the guard cells reached 20, which is significantly higher compared to the control (10–13), suggesting possible polyploid states or other morphogenetic changes. Analysis of genetic diversity with SSR markers revealed 28 alleles; the mutant lines showed new or altered alleles, especially when treated with 15 mg/l for 24 hours and with prolonged exposure to 10 mg/l for 48 hours. These changes show stable mutations, confirmed by the presence of unique allele combinations, which bring significant genetic diversity useful for breeding. Treating seeds with colchicine helps plants grow with higher productivity and genetic changes, expanding breeding possibilities and increasing pea yields without changing the level of polyploidy.
