Evolution, diversity and distribution of human Noroviruses:From single hosts to global patterns

Norovirus, a member of the Caliciviridae family, is a major cause of acute gastroenteritis (AGE) worldwide, affecting all age groups and causing substantial morbidity and mortality. Children in developing countries, the elderly, and immunocompromised individuals are particularly affected. Currently, no specific antivirals or vaccines are licensed for norovirus treatment or prevention. Therapeutic development is complicated by the virus's high diversity, with more than 10 genogroups (GI–GX) and approximately 50 genotypes, some of which, such as GII.4, continuously generate new antigenic variants. However, the emergence of norovirus diversity, the relationship between within-host and community-level evolution, and the association between viral diversity and treatment outcome remain incompletely understood. Therefore, this thesis aims to improve our understanding of norovirus evolution and epidemiology by addressing two main topics: norovirus diversity and evolution in different hosts, and the use of wastewater-based sequencing to monitor viral diversity and circulation at the population level.
