Bioinformatics Evaluation of the Association between LncRNA Linc00162 Gene and the Expression Alterations of Genes Involved in the PI3K/AKT/mTOR Signaling Pathway in Patients with Gastric Cancer

Introduction: Gastric cancer is one of the most common malignancies of the gastrointestinal tract and is associated with a high mortality rate.Recent evidence suggests that LncRNAs play a key role in regulating gene expression and cancer progression.In this study, the role of LncRNA PICSAR (LINC00162) in modulating the expression of genes involved in the PI3K/AKT/mTOR signaling pathway was investigated in patients with gastric cancer.In this study, gene expression data were collected from the TCGA database and gastric cancer-related datasets.Differentially expressed genes (DEGs) were identified using the DESeq2 and edgeR packages in R software version 4.2.2.Venn analysis was performed to identify genes shared between DEGs and co-expressed genes identified by PICSAR.Next, functional analysis of the pathways was performed using the online tool Enrichr and the KEGG databases, and significant pathways were identified based on adjP values < 0.05.Methods: Among 4640 differentially expressed genes and 988 co-expressed genes regulated by PICSAR, 163 common genes were identified.Pathway enrichment analysis revealed that the overlapping genes were enriched in multiple pathways, including PI3K/AKT/mTOR signaling, ECM-receptor interaction, and axon guidance.Also, ten key genes, including MMP7, KRT7, WNT10A, SEMA7A, and PMEPA1, showed the strongest correlation with PICSAR and the greatest expression changes in patients with gastric cancer.Results: This study indicates that LncRNA PICSAR may play an important role in the development and progression of gastric cancer by regulating the expression of key genes in the PI3K/AKT/mTOR pathway and other pathways associated with cancer progression.Conclusions: These findings indicate the high potential of PICSAR as a biomarker and therapeutic target for managing patients with gastric cancer.
