Green extraction and antioxidant validation of shallot peel waste

Shallot peels (Allium cepa L. var. aggregatum) are agricultural waste produced in large quantities during food processing. Improper disposal can lead to organic pollution and cause odor problems at landfill sites. Converting this biomass into high-value products offers a sustainable solution aligned with the principles of the circular economy. Shallot peels are rich in bioactive compounds, such as flavonoids and polyphenols, which have potential as antioxidants. However, studies on the antioxidant properties of shallots of the aggregatum variety using three antioxidant assay methods and the validation of these methods are still rarely reported. This study aimed to evaluate the antioxidant capacity of shallot skin waste using an environmentally friendly water-based decoction extraction technique and to rigorously validate its analytical performance. Antioxidant capacity was evaluated using three methods: DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2' -azino-bis(3ethylbenzothiazoline-6-sulfonic acid)), and FRAP (Ferric Reducing Antioxidant Power). Method validation was conducted according to international guidelines (ICH Q2(R1), AOAC SLV, ISO/IEC 17025:2017) to ensure accuracy, precision, reproducibility, and sensitivity. The extract demonstrated strong antioxidant activity with IC50 values of 5.85 ppm (DPPH), 2.73 ppm (ABTS), and 3.12 ppm (FRAP). Method validation showed high reproducibility (SD and CV <6%) and good sensitivity (low LoD and LoQ), indicating that this analytical method is consistent and reliable. This study indicates that aqueous extracts of shallot peels possess strong free radical scavenging and reducing capacities, supporting the principles of the circular economy and offering potential applications in the food, pharmaceutical, and cosmetic industries.
