Dual oxidized glycosaminoglycans crosslinked type II collagen hydrogel for in-situ full-thickness cartilage defect repair: in vitro and in vivo experiments

Hyaline articular cartilage (HAC) defects exhibit extremely limited self-repair capacity, while standard microfracture treatment tends to form inferior fibrocartilage. This study aimed to develop a dual oxidized glycosaminoglycan-crosslinked type II collagen (COL II) chondro-bionic hydrogel for in-situ repair of full-thickness articular cartilage defects. The hydrogel was prepared via Schiff base crosslinking at an optimized COL II/oxidized chondroitin sulfate/oxidized hyaluronic acid ratio of 80:15:5. It achieved rapid gelation within 5–10 min and exhibited mechanical properties consistent with native cartilage. In vitro , the hydrogel significantly promoted the proliferation and chondrogenic differentiation of bone marrow mesenchymal stem cells. In vivo , the hydrogel induced complete hyaline cartilage regeneration with biomechanical properties close to healthy cartilage at 6 months, and no systemic toxicity was detected. This hydrogel provides a safe, effective, and clinically translatable strategy for articular cartilage defect treatment
