Transdermal drug delivery systems (TDDS) have emerged as an effective alternative to conventional drug administration owing to their ability to bypass gastrointestinal degradation, minimize first-pass metabolism, and provide sustained therapeutic effects. The present study aimed to formulate and evaluate transdermal patches containing methanolic extracts of Gymnema sylvestre and Boerhavia diffusa for the management of diabetes mellitus. Hydroxypropyl methylcellulose (HPMC) was employed as the polymeric matrix, while polyethylene glycol (PEG) served as a plasticizer to enhance film flexibility and drug release characteristics. The herbal extracts were incorporated into transdermal patches using the solvent-casting method, and the prepared formulations were evaluated for physicochemical parameters including thickness, weight variation, folding endurance, moisture content, and moisture uptake. Drug–polymer compatibility was investigated using Fourier Transform Infrared (FTIR) spectroscopy. The prepared transdermal patches exhibited satisfactory physical appearance, flexibility, and stability. FTIR analysis indicated compatibility between the herbal extracts and excipients, with no evidence of significant chemical interaction. Among the developed formulations, selected patches demonstrated favorable moisture absorption, adequate mechanical strength, and sustained drug release characteristics with appreciable epidermal permeation over a period of approximately ten hours. These findings suggest that transdermal patches containing Gymnema sylvestre and Boerhavia diffusa may represent a promising herbal delivery platform for sustained antidiabetic therapy. However, further pharmacological, pharmacodynamic, and in vivo investigations are required to establish their therapeutic efficacy and long-term safety.
