Optimization of extraction technology and antioxidant activity of Crataegus songarica polysaccharide by response surface methodology

The experiment aimed to use the response surface method to optimize the ultrasonic extraction process, determine the optimal extraction process conditions for Crataegus songarica polysaccharide and study the antioxidant activity of Crataegus songarica polysaccharide. A Box-Behnken design was used to optimise a four-factor three-level test, the preliminary structural analysis of Crataegus songarica polysaccharide was analyzed by Fourier transform infrared spectroscopy, and in vitro antioxidant activity was studied. The results showed that the optimum extraction conditions for the extraction of Crataegus songarica polysaccharide were extraction time 41 min, extraction temperature 82 ℃, extraction power 210 W and liquid to material 17 mL/g. The yield of Crataegus songarica polysaccharide was 5.36%. Crataegus songarica polysaccharide has the characteristic absorption peaks of polysaccharide. Crataegus songarica polysaccharide has the ability to scavenge DPPH free radicals, ABTS free radicals, hydroxyl radicals and Ferric ion reducing power. The study indicates that the response surface methodology is feasible in optimising the ultrasonic extraction process design, and Crataegus songarica polysaccharide has good antioxidant activity and can be used as a natural antioxidant.
