scRNA-seq and VisiumHD of murine biomaterial-mediated fibrosis
Résumé fourni par la source
This repository contains scRNA-seq and VisiumHD datasets of a murine model of biomaterial-induced fibrosis. This model involves implanting polycaprolactone particles into a volumetric muscle loss injury in the murine quadriceps. Tissues were harvested at 6 weeks following muscle injury and biomaterial implantation. For the scRNA-seq dataset, cells were isolated from digested tissues and sorted using MACS CD45 microbeads. Cells were sequenced at a 1:1 ratio of CD45+ cells to CD45- cells using 10X Genomics Chromium 3' v3.1. For VisiumHD, tissues were fixed in 10% formalin for 48 hours and processed for paraffin embedding and tissue sectioning. VisiumHD was performed on one section per mouse (two sections total) using the manufacturer's protocol. Details on dataset processing are provided in the source publications. The processed datasets are provided as Seurat objects.
