Supplementary data includes Supplementary Materials and Methods, Supplementary Figures 1-6 with Legends and Supplementary table S1. Supplementary figures showing 1) characterization of isolated normal exocervical fibroblasts and 2) primary freshly isolated fibroblasts from cervical cancer outgrowth cultures, 3) isotype control stainings, 4) enhanced IL-23 but reduced IL-12 expression in cervical cancer-instructed mDC/NECF co-cultures, and elevated levels of IL23p19 and IL-12p40 but reduced IL-12p35 in cervical cancer-instructed mDC after co-cultivation with fibroblasts, 5) analysis of IL-6Ra expression of NECF and rhIL-6-induced IL-1b - and C/EBPb -expression in NECF, and 6) supernatants of cervical cancer-instructed mDC-NECF co-cultures only slightly induce Th17 but not Th1 differentiation, and neutralization of CCL20 in supernatants of fibroblasts did not affect IL-23 expression of DC and subsequent Th17 expansion. Supplementary table S1 shows the expression of maturation markers of LPS- and TNF-α-matured cervical cancer-instructed DC before and after co-cultures.
