Concurrent CDK9 and OxPhos inhibition inhibits growth of MCL cells. A, Parental and ibrutinib-resistant MCL cell lines were treated with the indicated doses of IACS-010579 for 72 hours. Cell proliferation was assessed using a colorimetric tetrazolium-based assay. The mean ± SEM is shown. *, P < 0.05 for JeKo-1 vs. JeKo-1R comparison. B, MCL cell lines were treated with IACS-010579 at the indicated doses for 24 hours (n = 3 tested in duplicates). Apoptosis was determined by flow cytometry using Annexin V–FITC staining. *, P < 0.05 and **, P < 0.01 compared with control. C, MCL cell lines were treated with IACS-010579 and/or AZD4573 as single agents or in combination at the indicated doses for 72 hours.
