Supplementary Figure S1: (A) Fraction of CD45-ve cells in the bone marrows isolated from PyMT NE+/+ (N=5) and PyMT NE-/- (N=5), used in Supplementary Figure S2. Gating strategies for the CD45+ and CD45- cells is shown in Supplementary Figure S10. (B) Representative images of elastin-stained lungs in FVB NE+/+ and FVB NE-/- mice at 12 weeks of age. Graphs to the right shows quantitation of the NE mediated cleavage of elastin in each set of genotypes. (C) Lung metastasis evaluation in the 4T1 model of breast cancer. 0.5X106 cells were injected by tail vein injection in BALB/c NE+/+ (N=9) and BALB/c NE-/- (N=5) mice. Lung metastases were evaluated 14 days post tail vein injections in both genotypes. (D) 0.5X106 4T1 murine TNBC cell lines was injected in the T4 mammary fat pad of BALB/c NE+/+ (N=6) and BALB/c NE-/- (N=5) mice. Tumor growth curves, as a function of time are shown. (E) Baseline neutrophils fraction per mg of mammary gland in non-tumor bearing FVB NE-/- mice compared to BALB/c NE-/- mice. (F) Quantitative assessment of ductal staining and branch staining of mammary glands (from PyMT NE+/+ and PyMT NE-/- mice), using the Aperio color deconvolution algorithm.
