Development of a Quercetin–Resveratrol Microemulsion Hydrogel Using an Isopropyl Myristate–Tween 80 System for Improved Healing of Diabetic Wounds

The present study aimed to develop and evaluate a microemulsion-based hydrogel incorporating quercetin and resveratrol using an isopropyl myristate–Tween 80–polyethylene glycol 400 system for enhanced topical delivery in diabetic wound healing. Solubility studies guided the selection of formulation components, and pseudo-ternary phase diagrams were constructed to identify the microemulsion region. The optimized microemulsion exhibited a droplet size of 118.4 ± 5.6 nm, low polydispersity index, and high entrapment efficiency (86.3 ± 2.1%). Incorporation into a Carbopol 934 hydrogel resulted in a stable, homogeneous formulation with suitable pH and rheological properties for topical application. In vitro drug release studies demonstrated sustained release of both quercetin (91.6 ± 3.1%) and resveratrol (94.3 ± 3.4%) over 24 hours, following Higuchi diffusion kinetics. The formulation showed enhanced permeation with flux values of 35.1 ± 1.02 µg/cm²/h and 37.3 ± 1.08 µg/cm²/h for quercetin and resveratrol, respectively. Antioxidant evaluation revealed superior DPPH radical scavenging activity compared to free drugs. Cell viability studies confirmed biocompatibility, while scratch assay results demonstrated significantly enhanced fibroblast migration (89.6 ± 3.2% wound closure at 48 h). The study concludes that the developed microemulsion-based hydrogel effectively enhances solubility, permeation, and biological activity of quercetin and resveratrol, making it a promising approach for diabetic wound management.
