Pharmacological modulation of circadian rhythms in rodent models of bipolar disorder: a scoping review protocol

Background: Circadian rhythms regulate diverse physiological and behavioral processes and are increasingly recognized as central to mood disorder pathophysiology, including bipolar disorder (BD). Disruptions in circadian timing affect sleep-wake cycles, hormonal regulation, and mood stability. Many pharmacological treatments for BD also influence circadian systems, suggesting that rhythm modulation may contribute to therapeutic efficacy. However, the extent and characteristics of preclinical research exploring this relationship remain unclear. Objective: This scoping review aims to map the extent, range, and characteristics of the literature examining pharmacological modulation of circadian rhythms in rodent models relevant to BD, specifically mouse (Mus musculus) and rat (Rattus Norvegicus). Methods: This review will follow the Joanna Briggs Institute methodology and report findings according to the PRISMA-ScR guidelines. Electronic databases including PubMed, Embase, Scopus, and Web of Science will be searched for peer‑reviewed studies evaluating pharmacological interventions that modulate circadian rhythms in rodent models exhibiting bipolar‑related phenotypes. Eligible studies will include experimental or quasi‑experimental designs assessing circadian outcomes alongside behavioral, physiological, or neurobiological measures. Study screening and data extraction will be performed independently by three reviewers using predefined criteria and a structured charting form. Results will be summarized descriptively in narrative and tabular formats. Conclusions: This review will provide an overview of current evidence, identify knowledge gaps, and guide future preclinical and clinical research aimed at assessing and developing circadian‑based therapeutic strategies for BD.
