From microbes to mind and actions: Uncovering the gut-brain connection in depression and its implications for human health and lifestyle

This study investigates associations between gut microbiome composition, depression, and lifestyle factors using 16S rRNA gene sequencing data from 7,472 participants in the Atlas Biomed Study, a large geographically diverse cohort spanning Western Europe, Eastern Europe, and Central Asia. Microbiome-wide association analyses were performed in independent discovery and replication cohorts, followed by meta-analysis, mediation analysis, Mendelian randomisation, and construction of microbiome risk scores (MRS) for depression. The study identified reduced gut microbial alpha diversity in individuals with a history of depression and replicated several previously reported depression-associated bacterial genera, including enrichment of Bacteroides, Tyzzerella, and Flavonifractor, alongside depletion of Coprococcus, Romboutsia, and Bifidobacterium. Four additional genera (Bacteroides pectinophilus group, Clostridium sensu stricto, Agathobacter, and Victivallis) were identified as novel taxa inversely associated with depression risk. Lifestyle factors such as diet, sleep, physical activity, smoking, and alcohol consumption were also examined as potential contributors to microbiome-mediated depression pathways. The work contributes to understanding the microbiome-gut-brain axis and highlights potential lifestyle and microbial targets relevant to depression risk and prevention.
