Smartphone-based digital imaging colorimetry of imeglimin using the novel chromaeus software

Imeglimin IMG is a recently approved antidiabetic agent used for the mangement of type 2 diabetes mellitus. Its routine quality control requires simple and green analytical methods. However, most reported analytical approaches rely on chromatographic or spectroscopic techniques that involve expensive instrumentation, time-consuming procedures, and high solvent consumption. In addition, existing smartphone-based and colorimetric methods lack standardized image processing workflows. Therefore, this work presents a smartphone assisted colorimetric method for the quantitative analysis of IMG. The method is based on the derivatization reaction between IMG and 1,2-naphthoquinone-4-sulfonate sodium (NQS) to form a colored product. Experimental parameters were optimized using a 2³ full factorial design. The novelty of this study arises from the application of the Chromaeus as a unified and standardized analytical workflow for color analysis of the derivatization product. The method showed linearity over the concentration range of 0.5–20 μg mL-1, with a limit of detection of 0.42 μg mL-1 and a limit of quantification of 1.28 μg mL-1. The method demonstrated accuracy (98–102%) and precision (RSD < 2%). The sustainability assessment using EPPI and RGB 12 confirmed excellent greenness. The developed method was applied to commercial tablets, with an average recovery of 99.18%.
