Membrane potential plays an important role during embryonic development, and the zebrafish serves as an effective model to study embryonic development. However, tools for reporting membrane potential in zebrafish remain limited. This study aimed to develop an in vivo optical methodology to investigate endogenous membrane potential during early zebrafish development. To achieve this goal, stable transgenic zebrafish lines expressing UAS-driven genetically encoded voltage indicators (GEVIs) were generated and validated. These fish lines were applied to the pax3a-marked mesoderm to observe membrane potential patterns. In vivo imaging confirmed the dynamic cellular membrane potential, indicating that GEVIs are functional in these fish lines. Longitudinal time-lapse imaging revealed oscillatory patterns within the somites. Overall, this study established transgenic UAS-ASAP zebrafish lines likely capable of reporting tissue-specific cell membrane potentials, providing a validated optical toolkit for future developmental studies.
