A Century of Pesticides use Reshaping a Lake Ecosystem (Metabarcoding data - Saint-André lake, France)

Metabarcoding data obtained from 20 slices of a sediment core from lake Saint-André (France) ranging from 2016 to 1879. DNA was extracted for each slice. DNA extracts were amplified with an eukaryotic targeting primer pair (Euka02, TTTGTCTGSTTAATTSCG- CACAGACCTGTTATTGC, Guardiola et al. (2015)) which targets a short 18S rDNA fragment (~123bp). Each DNA extract has been amplified 8 times. Multiplexing was done by adding a unique 8bp oligonucleotide tag to the 5′ of the forward and reverse primers. PCR products were all pooled for library preparation. The library was sequenced twice on a HiSeq 2500 Illumina sequencer (pair-end, 2x125bp).
