SPERM CREATINE KINASE ACTIVITY: ISIT A CRITERTA FOR FERTILIZATION POTENTTAL OF SPERM?

Abstract The aim of this study is to examine the valuc of sperm creatine kinase to predict fertilizing potential of sperm. 32 patients (13 male factor/19 famele factor) who attended to IVF Center were entered in this study. Pellets which were oblained by centrifugation of ligueficd semen samples were used for creatine phosphokinase MB (CPK- MB) isoform determination alter intracellular enzymes were exiracted. CPK-MB enzyme activity was determined using a kinetic specitrophotometric Diasis Diagnostic kit which works on basis of NADPH absorbtion at 340 nm. CPK-MB levels were given as activity per mg of protein (Ü/L/mg prot.) Levels of CPK-MB were stalisticaliy lower in 18 patients (mean 18.7144.62 Ü/L/mg prot.) with positive in vit- ro ferlilization than in 14 patients (mcan 30.32-44.32 Ü/L/mg prot.) without fertilization. In 4 of 13 patients with ma- le infertility, fertilization was obtained and mean CPK-MB level was 24.2342.37 Ü/L/mg prot., while it was 30.9813.44 Ü/L/mg prot. in the other 9 patients. The difference was significant. CPK-MB levels were significantly lower in 19 normozoospermic patients (mcan 28.9144.45 Ü/L/mg prot.) (p<0.05). With respect to sperm morpho- logy, there was also statistically significani difference between groups with fertilization positive and negative in both normozoospermic and oligozoospermic patients (p<0.05). Determination ol sperm CPK-MB activity may be helpful to choose the patients with male infertility who may benefit from artificial insemination technigues. But, studies are nceded to delermine if it is superior to sperm morp- hology.
