Estimating Total Estrogens in Human Serum Samples Using Competitive ELISA

Estrogens are a group of hormones that play an important role in normal sexual and reproductive development. In females, most of the estrogen hormones are synthesized by the ovary, although small amounts are also synthesized by the adrenal glands and fat cells. Estimating total estrogens in blood aids in diagnosing sex steroid-related conditions such as deficiencies of enzymes like aromatase and 17 alpha-hydroxylase and conditions like premature or delayed puberty. In postmenopausal women, and in older men with lower accuracy, blood estrogen levels can predict fracture risk. Estrogen is also measured to investigate the outcomes of hormone replacement therapy in postmenopausal women. One of the techniques commonly used for the estimation of estrogens in blood is enzyme-linked immunosorbent assay (ELISA). This immunoassay is based on antibodies to detect a target antigen using highly specific antibody-antigen interactions. There are different types of ELISA; the authors of this chapter use competitive ELISA.
