Effects of exposure to Fe3O4-PEG-PLGA nanoparticles on the lung of wild type and Nrf2-KO mice

[Background] Recently, various kinds of bionanomaterials have been developed for the purpose of practical use in medical area or other industrial areas. Fe3O4-PEG-PLGA nanoparticles have been expected to be used in cancer diagnosis or hyperthermia treatment with near infrared radiation. The increase in Fe3O4-PEG-PLGA production might lead to increase in population exposed during manufacturing or using process. However, safety evaluation of Fe3O4-PEG-PLGA nanoparticles is still insufficient. The present study aims to investigate pulmonary effects of Fe3O4-PEG-PLGA nanoparticles and the role of Nrf2. [Methods] Wild type and Nrf2-/- were exposed once to Fe3O4-PEG-PLGA nanoparticles at 0, 10, 30 µg/mouse by pharyngeal aspiration. At 14 days after the exposure, bronchoalveolar lavage BALF was collected to quantify protein level and the number of inflammatory cells in it.
