Simultaneous determination of free/conjugated phytosterols and phytosterol oxidation products in vegetable oils by liquid-liquid extraction coupled with SPE-GC-MS

A method for the simultaneous determination of free/conjugated phytosterols and phytosterol oxidation products in vegetable oils was developed and validated in this study. Free phytosterols (FPS) and free phytosterol oxidation products (FPOPs) were directly separated and quantified by SPE-GC-MS, while conjugated phytosterols (CPS)/conjugated phytosterol oxidation products (CPOPs) were indirectly determined by subtracting FPS/FPOPs from total phytosterols/total phytosterol oxidation products obtained after cold saponification, liquid-liquid extraction, and SPE-GC-MS analysis. The limits of detection for FPS, FPOPs, CPS, and CPOPs were less than or equal to 0.12, 0.08, 0.10, and 0.08 μg/g, respectively, while the lower limits of quantification were less than or equal to 0.40, 0.26, 0.33, and 0.26 μg/g. Recoveries of spiked FPS, CPS, FPOPs, and CPOPs at low, medium, and high concentrations ranged from 75.84 % to 100.85 %, with within-day and between-day precision below 10.39 % in relative standard deviations. This method offers a simple, rapid, and novel way to simultaneously analyze different FPS/CPS and FPOPs/CPOPs in vegetable oils. • Various free and combined phytosterols (PS) were quantified. • Various free and combined phytosterol oxidation products (POPs) were quantified. • A method for simultaneous determination of free/combined PS and POPs was developed. • Combined PS and POPs were indirectly detected.
