Early intervention of IL-5 mab attenuates EMT process and airway remodelling in asthmatic mice

It has been identified that application of IL-5 mab attenuates the symptoms and asthma exacerbation, but whether it can improve airway remodelling remains unclear. We used ovalbumin (OVA) sensitization/challenge to build chronic asthma model and IL-5 mab was administrated twice a week for the lattar 4 weeks or 1 week. Each treatment group was divided into two groups including high (60mg/kg) and low (20mg/kg) dosage respectively. After the last OVA challenge, we performed spirometry and AHR measurement. And we collected the lung tissue to evaluate the expression of EMT markers and subepithelial fibers formation as well as collagen deposition associated with airway remodelling, and to evaluate the level of cytokines in bronchoalveolar lavage fluid (BALF). When treated by IL-5 mab, early intervention obviously improved the large and small airway function compared with asthma mice, but late intervention only mitigated small airway function in high dosage group. Early and late intervention attenuated AHR. High dosage of IL-5 mab administration at early period reduced the level of MMP9 significantly. And early intervention showed a significant decreasing in the area of Masson’s trichrome and it ameliorated subepithelial fibers formation and collagen I deposition, as well, the expression of E-cadherin was increased with reduction of N-cadherin and α-SMA. IL-5 mab early intervention is better than late intervention in improving lung function. Small airway function is improved earlier than large after the application of IL-5 mab. High dosage administration as soon as possible can reverse EMT process and asthma airway remodelling.
