Organocatalysis, utilizing small organic molecules to drive asymmetric transformations, has emerged as a powerful and versatile platform for synthesizing valuable chiral compounds. This chapter systematically reviews privileged organocatalysts, classifying them according to five distinct activation modes respectively involving imine/iminium intermediate formation, Brønsted acid–base activation, Lewis acid–base coordination, cation–anion interactions, and dioxirane-mediated transformations. The review also pays special attention to structural diversity of the organocatalysts and their applications in advancing stereoselective synthetic methodologies. Finally, the review addresses persistent challenges and tentatively outlines future research directions in this rapidly evolving field.
