Advances in the diagnosis and treatment of marginal zone lymphomas

Marginal zone lymphomas (MZL) are a heterogeneous group of indolent B-cell non-Hodgkin lymphomas encompassing extranodal, nodal, and splenic subtypes. Over the past decade, significant progress has been achieved in understanding their biology, refining diagnostic approaches, and expanding therapeutic options. In addition to established treatments such as antibiotic eradication therapy, radiotherapy, and chemoimmunotherapy, novel targeted agents — including immunomodulatory drugs, covalent and non-covalent Bruton tyrosine kinase inhibitors (BTKi), CAR-T cell therapies, bispecific antibodies, and antibody–drug conjugates (ADCs) — are increasingly being incorporated into clinical practice. This review summarizes current knowledge on the epidemiology, biology, treatment strategies, and emerging therapeutic challenges associated with MZL.
